Exploring the Potential of Atlantic Mesopelagic Species Processed on Board Commercial Fishing Vessels as a Source of Dietary Lipids.

This study investigates the use of untapped mesopelagic species as a source of long-chain polyunsaturated omega-3 fatty acids (LC n-3 PUFAs) to meet the growing demand. The challenges faced by commercial fishing vessels, such as varying catch rates and species distribution affecting lipid levels, are addressed. Marine oils were produced post-catch using thermal separation and enzymatic hydrolysis during four commercial cruises, screening approximately 20,000 kg of mixed mesopelagic species. Maurolicus muelleri and Benthosema glaciale were the dominant species in the catch, while krill was the primary bycatch. The lipid composition varied, with B. glaciale having a higher prevalence of wax esters, while triacylglycerols and phospholipids were more predominant in the other species. LC n-3 PUFAs ranged from 19% to 44% of lipids, with an average EPA + DHA content of 202 mg/g of oil. Both processing methods achieved oil recoveries of over 90%. Estimates indicate that the mesopelagic biomass in the Northeast Atlantic could supply annual recommended levels of EPA + DHA to 1.5 million people, promoting healthy heart and brain functions. These findings offer valuable insights for considering mesopelagic species as a potential source of dietary marine lipids, laying the groundwork for further research and innovation in processing and obtaining valuable compounds from such species.

Genome-scale metabolic modeling of Thraustochytrium sp. RT2316-16: Effects of nutrients on metabolism.

Marine thraustochytrids produce metabolically important lipids such as the long-chain omega-3 polyunsaturated fatty acids, carotenoids, and sterols. The growth and lipid production in thraustochytrids depends on the composition of the culture medium that often contains yeast extract as a source of amino acids. This work discusses the effects of individual amino acids provided in the culture medium as the only source of nitrogen, on the production of biomass and lipids by the thraustochytrid Thraustochytrium sp. RT2316-16. A reconstructed metabolic network based on the annotated genome of RT2316-16 in combination with flux balance analysis was used to explain the observed growth and consumption of the nutrients. The culture kinetic parameters estimated from the experimental data were used to constrain the flux via the nutrient consumption rates and the specific growth rate of the triacylglycerol-free biomass in the genome-scale metabolic model (GEM) to predict the specific rate of ATP production for cell maintenance. A relationship was identified between the specific rate of ATP production for maintenance and the specific rate of glucose consumption. The GEM and the derived relationship for the production of ATP for maintenance were used in linear optimization problems, to successfully predict the specific growth rate of RT2316-16 in different experimental conditions.

Unwrapping the (glyco-)lipidome in the microalgae Microchloropsis gaditana: Effects of eco-friendly extraction methods

Microalgae have recently captivated worldwide attention because of their extraordinary lipid composition, which includes essential omega-3 and omega-6 long-chain polyunsaturated fatty acids (LC-PUFA), neutral lipids, and more complex lipids (glycolipids and phospholipids), as well as eicosanoids. To explore this further, we present a novel (glyco-)lipidomics strategy, including a green chemistry extraction approach, liquid chromatography high-resolution mass spectrometry (LC-HRMS) lipid profiling, and a comprehensive data evaluation workflow. Using untargeted lipidomics based on LC-HRMS approaches, we studied the lipid composition up to the molecular species level of the microalgae Microchloropsis gaditana (formerly Nannochloropsis gaditana), focusing on the distribution of saturated, monounsaturated, and polyunsaturated fatty acids in different lipid classes. Our analysis successfully identified 520 lipid molecular species from 18 lipid classes. Regarding fatty acid composition, our primary focus was on elucidating the distribution of LC-PUFA across the different microalgal lipid classes. This particular strategy holds potential as it may unravel the use of microalgal lipids as innovative sources of food ingredients. Our findings show that LC-PUFA are relatively elevated in membrane lipids, in which cholesterol esters (ChE) have the highest PUFA share (94 %), followed by polar membrane lipids (glycolipids and phospholipids), with phosphatidylethanolamines (PE) up to 67 %, phosphatidylglycerols (PG) up to 47 %, and monogalactosyldiacylglycerols (MGDG) up to 47 % PUFA content. This highlights the high percentage of PUFA in the structural parts of microalgae and the importance of these lipid classes as a reservoir for PUFA. Finally, we also compared a classical Folch extraction method with environmentally friendly ultrasound-assisted extraction approaches and enzymatic pre-treatments to investigate the lipid composition and enrichment of valuable lipid classes. Thus, this extensive analysis of the (glyco-)lipidome of Microchloropsis gaditana provides valuable insights into the existing knowledge about this microalgae species, enhancing our understanding of the complex lipid composition and opening up new opportunities for potential applications in the biotechnology and food industry.

Muscle biopsy long-chain omega-3 polyunsaturated fatty acid compositions, IMF and FMP in Australian pasture-based Bowen Genetics Forest Pastoral Angus, Hereford, and Wagyu Beef Cattle

BackgroundWe investigated breed and gender variations in the compositions of long-chain (≥ C20) omega-3 polyunsaturated fatty acids (LC omega-3 PUFA), fat melting point (FMP) and intramuscular fat (IMF) contents in biopsy samples of the M. longissimus dorsi muscle of grazing beef cattle. The hypothesis that biopsy compositions of health-beneficial LC omega-3 PUFA, FMP and IMF in a pasture-based production system will vary with breed, was tested. Muscle biopsies were taken from 127 yearling pasture-based Angus, Hereford, and Wagyu heifers and young bulls exclusive to the Australian Bowen Genetics Forest Pastoral breeding stud averaging 12 ± 2.43 months of age and under the same management routine.ResultsBreed had a significant influence on IMF, FMP, and the compositions of oleic acid, α-linolenic acid (ALA), eicosapentaenoic (EPA), docosahexaenoic (DHA), docosapentaenoic (DPA), and total EPA + DHA + DPA in the M. longissimus dorsi muscle biopsies (P ≤ 0.03). The Wagyu breed had the highest (11.1%) and Hereford the lowest (5.9%) IMF (P = 0.03). The reverse trend was observed in FMP values where the Hereford breed had the highest (55 °C), Angus intermediate (46.5 °C), and Wagyu the lowest (33 °C) FMP. The Wagyu and Angus breeds had similar oleic fatty acid (18:1n-9) content, while the Hereford breed had the lowest (P < 0.01). The highest ALA, DPA, total EPA + DHA, total EPA + DHA + DPA and total ALA + EPA + DHA + DPA contents were detected in the Wagyu breed (P ≤ 0.03). The Hereford had similar EPA and DPA contents to the Angus (P ≥ 0.46). Total EPA + DHA + DPA contents in Wagyu, Angus, and Hereford were 28.8, 21.5, and 22.1 mg/100g tissue (P = 0.01), respectively. Sex was an important source of variation that influenced LC omega-3 PUFA composition, FMP and IMF, where yearling heifers had higher IMF (11.9% vs 5.3%), lower FMP (33°C vs 37°C), and higher LC omega-3 PUFA than bulls.ConclusionAll the results taken together indicate that the Wagyu breed at 28.8 mg/100g tissue, was the closest to meeting the Australia and New Zealand recommended source level threshold of 30 mg/100g tissue of health-beneficial ≥ C20 omega-3 FA content. Since gender was a significant determinant of LC omega-3 PUFA composition, IMF content and FMP, it should be factored into enhancement strategies of healthy meat eating quality traits in grazing cattle. These findings also suggest that the Bowen Genetics Forest Pastoral beef cattle studs are important sources of LC omega-3 PUFA that can be used to cover the deficit in these health claimable fatty acids in Western diets.

The Maternal Omega-3 Long-Chain Polyunsaturated Fatty Acid Concentration in Early Pregnancy and Infant Neurodevelopment: The ECLIPSES Study.

Omega-3 Long-Chain Polyunsaturated Fatty Acids (n-3 LCPUFAs) play a key role in early neurodevelopment, but evidence from observational and clinical studies remains inconsistent. This study investigates the association between maternal n-3 LCPUFA, Docosahexaenoic Acid (DHA), and eicosapentaenoic acid (EPA) concentrations during pregnancy and infant development functioning at 40 days. This study includes 348 mother-infant pairs. Maternal serum concentrations were assessed in the first and third trimesters alongside sociodemographic, clinical, nutritional, psychological, and obstetrical data. At 40 days, the Bayley Scales of Infant and Toddler Development, Third Edition (BSID-III) was administered. An adjusted analysis revealed that lower first-trimester n-3 LCPUFA and DHA concentrations are associated with better infant motor development. These results underscore the potential significance of the maternal n-3 LCPUFA status in early pregnancy for influencing fetal neurodevelopment. However, the complexity of these associations necessitates further investigation, emphasizing the urgent need for additional studies to comprehensively elucidate the nuanced interplay between the maternal n-3 LCPUFA status and infant neurodevelopment.

Omega-3 long-chain polyunsaturated fatty acids and their bioactive lipids: A strategy to improve resistance to respiratory tract infectious diseases in the elderly?

Age-related changes in organ function, immune dysregulation, and the effects of senescence explain in large part the high prevalence of infections, including respiratory tract infections in older persons. Poor nutritional status in many older persons increases susceptibility to infection and worsens prognosis. Interestingly, there is an association between the amount of saturated fats in the diet and the rate of community-acquired pneumonia. Polyunsaturated fatty acids, particularly omega-3 long chain polyunsaturated fatty acids (ω-3 LC-PUFAs) including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), have well-known anti-inflammatory, immunomodulatory, and antimicrobial effects, which may, in theory, be largely induced by PUFAs-derived lipids such as specialized pro-resolving mediators (SPMs). In adults, preliminary results of studies show that ω-3 LC-PUFAs supplementation can lead to SPM generation. SPMs have a crucial role in the resolution of inflammation, a factor relevant to survival from infection independent of the pathogen’s virulence. Moreover, the immune system of older adults appears to be more sensitive to ω-3 PUFAs. This review explores the effects of ω-3 LC-PUFAs, and PUFA bioactive lipid-derived SPMs in respiratory tract infections and the possible relevance of these data to infectious disease outcomes in the older population. The hypothesis that PUFAs have beneficial effects via SPM generation will need to be confirmed by animal experiments and patient-derived data.

Optimal omegas - barriers and novel methods to narrow omega-3 gaps. A narrative review.

Dietary intakes of omega-3 long chain polyunsaturated fatty acids (O3LC-PUFAs) such as eicosapentaenoic and docosahexaenoic acid are central to development and health across the life course. O3LC-PUFAs have been linked to neurological development, maternal and child health and the etiology of certain non-communicable diseases including age-related cognitive decline, cardiovascular disease, and diabetes. However, dietary inadequacies exist in the United Kingdom and on a wider global scale. One predominant dietary source of O3LC-PUFAs is fish and fish oils. However, growing concerns about overfishing, oceanic contaminants such as dioxins and microplastics and the trend towards plant-based diets appear to be acting as cumulative barriers to O3LC-PUFAs from these food sources. Microalgae are an alternative provider of O3LC-PUFA-rich oils. The delivery of these into food systems is gaining interest. The present narrative review aims to discuss the present barriers to obtaining suitable levels of O3LC-PUFAs for health and wellbeing. It then discusses potential ways forward focusing on innovative delivery methods to utilize O3LC-PUFA-rich oils including the use of fortification strategies, bioengineered plants, microencapsulation, and microalgae.

Diets that do not let benthivorous salmonid fish, Thymallus baicalensis, realize their potential for accumulating large content of omega-3 PUFAs in muscle tissue

Salmoniformes are valuable commercial fish that are high in physiologically important omega-3 long-chain polyunsaturated fatty acids (LC-PUFAs), namely eicosapentaenoic (EPA; 20:5n-3) and docosahexaenoic (DHA; 22:6n-3) ones. The content of EPA and DHA and, as a result, the quality of fish for consumers depend on the fish diet. We studied the fatty acid (FA) content and composition of grayling, Thymallus baicalensis, inhabiting six rivers, which differed in food supply for fish. The diet of fish was studied using fatty acid markers in adipose tissue and stomach content. In two rivers, grayling's diet was of aquatic origin, while in four rivers, terrestrial invertebrates constituted a high proportion of the fish diet. The study was conducted to test the following question: How do diets with varying amounts of terrestrial-based food influence the content of EPA and DHA, as well as the fatty acid composition, in various tissues of grayling? Grayling inhabiting shaded rivers had lower nutritional value as a source of LC-PUFAs for consumers compared to those in unshaded rivers. This was reflected in the diets of grayling in shaded rivers, which included more prey relying on terrestrial sources of FAs compared to unshaded rivers, where dietary items relied on autochthonous production. Furthermore, these different diets greatly affected the content of EPA and DHA in muscle and adipose tissue but demonstrated a less significant impact on PUFA content in the brain. These findings are of interest because they suggest that the nutritional value of grayling tissues, consumed by predators, including humans, depends on source material within a river and adjacent terrestrial habitat.

Docosahexaenoic Acid (DHA) Down-regulates Cellular, and Exosomal Overexpressed Mammalian Target of Rapamycin (mTOR) in Breast Cancer Cells Under Hypoxic Condition

Background: The omega-3 long-chain polyunsaturated fatty acid, docosahexaenoic acid (DHA), shows anti-proliferative effects in cancer cell lines and animal models. The mammalian target of rapamycin (mTOR) is one of the regulators for the proliferation and survival of cancer cells. This study focused on the effect of DHA on cellular and exosomal expression of mTOR and related tumor-suppressor microRNAs (miRs) in triple-positive (BT-474) and triple-negative (MDA-MB-231) breast cancer (BC) cell lines. Methods: BT-474 and MDA-MB-231 cells were treated with 100 μM DHA under hypoxic and normoxic conditions for 24 hours. The exosomes were isolated by ultracentrifuge and determined by electron microscopy and CD9, CD63, and CD81 immunoblotting. cDNAs from cellular and exosomal total RNA were used for evaluation of the expression of mTOR and related tumor-suppressor miRs, miR-101 and miR-214, by quantitative real-time PCR. Results: We demonstrated that DHA significantly decreased cellular and exosomal expression of mTOR in both normoxic and hypoxic conditions for both cell lines. Consistently, DHA caused significantly increased expression of miR-214 in all treated groups. However, altered expression of miR-101 showed different patterns in cells and exosomes. Conclusion: According to the beneficial effect of DHA in decreasing the expression of a master regulator for the proliferation of cancer cells, mTOR, in part by increased expression of miR-214, it could be used as a supplementary therapy in BC treatment. Also, miRNA replacement therapy would be useful by suppressing the expression of mTOR in BC treatment.

Influence of Dietary Seafood on the Prevention of Ocular Disorders: A Clinical Analysis

Age-related macular degeneration, retinal dysfunction, dry eye syndromes, cataract formation, and the prevalence of other vision defects are directly related to nutrients in the diet. People consuming seafood are less prone to several vision disorders. The present study was designed to validate the view. In this study, 712 visitors to tertiary eye care for eye check-ups were interviewed using a standard questionnaire, and from them, 264 persons were identified with some ocular defects. The identified cases were further screened based on their dietary habits with seafood. The isolated cases were clinically examined, and the development of ocular defects such as macular edema, optic neuropathy, myopia, double vision, dry eye syndrome, and color blindness was recorded. The results show that the respondents taking one or another type of three fishes regularly have a low impact on visual disorders. Respondents with fish intake above 500 g/week were less affected by all five ocular disabilities tested when compared with occasional users. Among the total respondents, the percentage of visual defects was seen in 62.12% in seafood users, but among the non-seafood users, the presence of visual defects was 89.6%. Statistical analysis using the Pearson population correlation coefficient, ρ (rho), further confirmed the relationship between eye wellness and a seafood diet. The omega-3 fatty acids linked with vision were also high in the fish consumed by the people. This study confirms that a regular seafood diet is good for eye health and can prevent the epidemics of several eye diseases. As fish intake contains a good amount of long-chain polyunsaturated omega-3 fatty acids, people who consume fish in their diet are less susceptible to advanced macular degeneration and other vision impairments responsible for age-related blindness.

Exploring Omega-3's Impact on the Expression of Bone-Related Genes in Meagre (Argyrosomus regius).

Dietary supplementation with Omega-3 fatty acids seems to promote skeletal health. Therefore, their consumption at imbalanced or excessive levels has offered less beneficial or even prejudicial effects. Fish produced in aquaculture regimes are prone to develop abnormal skeletons. Although larval cultures are usually fed with diets supplemented with Omega-3 Long Chain Polyunsaturated fatty acids (LC-PUFAs), the lack of knowledge about the optimal requirements for fatty acids or about their impact on mechanisms that regulate skeletal development has impeded the design of diets that could improve bone formation during larval stages when the majority of skeletal anomalies appear. In this study, Argyrosomus regius larvae were fed different levels of Omega-3s (2.6% and 3.6% DW on diet) compared to a commercial diet. At 28 days after hatching (DAH), their transcriptomes were analyzed to study the modulation exerted in gene expression dynamics during larval development and identify impacted genes that can contribute to skeletal formation. Mainly, both levels of supplementation modulated bone-cell proliferation, the synthesis of bone components such as the extracellular matrix, and molecules involved in the interaction and signaling between bone components or in important cellular processes. The 2.6% level impacted several genes related to cartilage development, denoting a special impact on endochondral ossification, delaying this process. However, the 3.6% level seemed to accelerate this process by enhancing skeletal development. These results offered important insights into the impact of dietary Omega-3 LC-PUFAs on genes involved in the main molecular mechanism and cellular processes involved in skeletal development.

Isolation of intake mediated effects demonstrate that the phenomic benefits of dietary omega-3 are nominal to Atlantic salmon when reared in a challenging (hypoxic) environment

The present study was designed to examine whether challenging environmental conditions (such as hypoxia) influence a fish's response to diet by changing responses to nutrient utilisation or down-regulation of appetite. To examine this, post-smolt Atlantic salmon (138.8 ± 0.65 g/fish; mean ± SD) were fed sub- (2.6% TFA; Ln3) and optimal (6.0% TFA; Hn3) levels of long-chain polyunsaturated omega-3 fatty acids (n-3 LC-PUFA) when maintained under either normoxic (8.1 ± 0.6 mg/L) or hypoxic (6.8 ± 0.5 mg/L) environmental conditions. To control for the anticipated effects of hypoxia on appetite and food intake, two additional treatments included the two diets that were pair-fed to the same intake levels as in the hypoxic treatments. The reduction in dissolved oxygen was accompanied by an increase in dissolved CO2 and decline in seawater pH, with no change in total alkalinity. After 138 days of the study the fish increased their average weight to between 616 and 720 g/fish subject to treatment. Hypoxia had a significant negative impact on final weight and weight gain, and these were the result primarily of effects on feed intake. No effects of treatments on feed conversion were observed. Significant diet x oxygen interactions were observed on whole-body lipid levels, but no other proximate compositional parameters. Sub-clinical effects of hypoxia and ration on several plasma chemistry parameters were observed, but few responses were diet related. Hypoxia affected plasma total protein, cholesterol, and red blood cell counts. Diet digestibility was typically not affected by hypoxia or ration, though a range of diet related effects were evident. Nutrient retention data showed evidence of elongation and desaturation of dietary short-chain PUFA, with clear effects of diet affecting the deposition of both omega-6 and omega-3 fatty acids. This study builds on findings from earlier studies, but crucially now include controls for the feed intake effects to clarify the roles of hypoxia on the nutrient and intake mediated effects. As a result of these controls, it is now clearer that many of the phenomic responses to long-chain omega-3 by Atlantic salmon are nominal and are largely unaffected by challenging environmental conditions like hypoxia. The inclusion of a series of pair-fed treatments, matching the feed intake levels of corresponding hypoxic treatments, shows that the effects of n-3 LC-PUFA appear to be largely intake mediated effects for Atlantic salmon post-smolts.

The role of nutrition in the prevention of cognitive decline.

Dementia is a growing concern and underscores the urgent need for effective preventive measures targeting modifiable risk factors. Nutrition is a key player in the onset and progression of inflammation and cognitive decline. This review provides a comprehensive overview of the effects of different dietary patterns, vitamins and nutrients for preventing cognitive decline, mainly among healthy individuals and those with mild cognitive impairment. The Mediterranean diet, omega-3 long-chain polyunsaturated fatty acids and B vitamins are the most investigated, with evidence supporting protection against cognitive decline among older adults varying across studies. More recent interventions examined in this review, such as MIND Diet, are promising with positive results, but further research is needed to conclusively establish their efficacy. It is also crucial to consider complete lifestyle as physical activity for preventing cognitive decline. Definitive conclusions are difficult to draw. Future studies should adopt a comprehensive approach and focus on multinutrient strategies and whole diets.

Compositional and metabolic responses of European seabass (Dicentrarchus labrax) to a dietary gradient of long-chain (≥ C20) polyunsaturated fatty acids

Decrease in the inclusion of fish oil (FO) in current aquafeed formulations reduces the availability of omega-3 (n-3) long-chain polyunsaturated fatty acids (LC-PUFA) to marine fish like the European seabass Dicentrarchus labrax. Here we investigated the compositional and physiological responses of European seabass to a dietary gradient of n-3 LC-PUFA. Six isoenergetic and isonitrogenous diets (D1-D6) were fed to triplicate groups of juvenile seabass (23 g), containing blends of FO and vegetable oil (VO) that resulted in a gradient of n-3 LC-PUFA varying from 0.23 to 3.40% of diet as fed. Sampling was carried out after four months feeding and biochemical composition, as well as lipid metabolic gene expression in liver and mid-intestine analysed. The only effect of diet on proximate composition was a decrease in hepatic lipid content as dietary n-3 LC-PUFA increase whereas, in contrast, fatty acid (FA) compositions of both tissues were substantially modified by diet. Concentrations of LC-PUFA biosynthesis intermediates decreased in both tissues as dietary n-3 LC-PUFA increased with some preference detected towards n-6 FA. Two genes, namely fads2 and igf1, were moderately influenced by dietary VO in liver while genes involved in lipogenesis (srebp1, srebp2 and fas) and LC-PUFA biosynthesis (fads2 and elovl5) were markedly down-regulated, in mid-intestine of seabass in response to increasing dietary n-3 LC-PUFA. The expression of genes of β-oxidation (ppara, cpt1a and fabp1) did not vary among treatment. Collectively, our data suggest that the nutritional regulation of key lipid metabolic genes in European seabass was more pronounced in mid-intestine than in liver. Overall, when compared to a previous study of similar design in gilthead seabream (Sparus aurata), the European seabass seems less sensitive to a dietary gradient of vegetable oil.

Differential expressions of FASN, SCD, and FABP4 genes in the ribeye muscle of omega-3 oil-supplemented Tattykeel Australian White lambs

BackgroundThe concept of the functional nutritional value of health-beneficial omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) is becoming a phenomenon among red meat consumers globally. This study examined the expressions of three lipogenic genes (fatty acid binding protein 4, FABP4, fatty acid synthase, FASN; and stearoyl-CoA desaturase, SCD) in the ribeye (Longissimus thoracis et lumborum) muscle of Tattykeel Australian White (TAW) lambs fed fortified omega-3 diets and correlations with fatty acids. To answer the research question, “are there differences in the expression of lipogenic genes between control, MSM whole grain and omega-3 supplemented lambs?”, we tested the hypothesis that fortification of lamb diets with omega-3 will lead to a down-regulation of lipogenic genes. Seventy-five six-month old TAW lambs were randomly allocated to the (1) omega-3 oil-fortified grain pellets, (2) unfortified grain pellets (control) or (3) unfortified MSM whole grain pellets diet supplements to generate three treatments of 25 lambs each. The feeding trial lasted 47 days.ResultsFrom the Kruskal-Wallis test, the results showed a striking disparity in lipogenic gene expression between the three dietary treatments in which the FABP4 gene was significantly up-regulated by 3-folds in the muscles of lambs fed MSM Milling (MSM) whole grain diet compared to the omega-3 and control diets. A negative correlation was observed between FASN gene expression and intramuscular fat (IMF), eicosapentaenoic acid (EPA), total polyunsaturated fatty acids (PUFA), omega-6 polyunsaturated fatty acids (n-6 PUFA) and monounsaturated fatty acids (MUFA). The FABP4 gene expression was positively correlated (P < 0.05) with EPA and docosahexaenoic acid (DHA).ConclusionTaken together, this study’s results suggest that FABP4 and FASN genes perform an important role in the biosynthesis of fatty acids in the ribeye muscle of TAW lambs, and supplementary diet composition is an important factor influencing their expressions.

Therapeutic Benefits of Tuna Oil by In Vitro and In Vivo Studies Using a Rat Model of Acetic Acid-Induced Ulcerative Colitis.

Ulcerative colitis (UC), an inflammation of the colon lining, represents the main form of inflammatory bowel disease IBD. Nutritional therapy is extremely important in the management of ulcerative colitis. Fish oil contains long-chain omega-3 polyunsaturated fatty acids, which have beneficial effects on health, including anti-inflammatory effects. This study aims to investigate the benefits of bluefin tuna oil extracted by the Soxhlet method in vitro by determining the anti-radical and anti-inflammatory activities and in vivo by evaluating the preventive and curative effects. The experiments were carried out using two doses of oil (100 and 260 mg/kg) and glutamine (400 and 1000 mg/kg) on the acetic acid-induced UC model. UC has been induced in Wistar rats by intrarectal administration of a single dose of 1 mL acetic acid (5% v/v in distilled water). The obtained results indicate that tuna oil and glutamine have a significant anti-free radical effect. Tuna oil has a marked anti-inflammatory power based on membrane stabilization and inhibiting protein denaturation. The reduction of various UC parameters, such as weight loss, disease activity score DAS, and colonic ulceration in rats pre-treated with tuna oil and glutamine, demonstrate that these treatments have a significant effect on UC. Total glutathione GSH, superoxide dismutase SOD, and catalase activities are significantly restored in the tuna oil and glutamine groups, while lipid peroxidation has been markedly reduced.

Lipidomic studies reveal two specific circulating phosphatidylcholines as surrogate biomarkers of the omega-3 index

Optimal dietary intake of omega-3 long-chain polyunsaturated fatty acids (n3-LCPUFAs) is critical to human health across the lifespan. However, omega-3 index (O3I) determination is not routinely assessed due to complicated procedures for n3-LCPUFA analysis from the phospholipid (PL) fraction of erythrocytes. Herein, a high-throughput method for lipidomics based on multisegment injection-nonaqueous capillary electrophoresis-mass spectrometry was applied to identify circulating PLs as surrogate biomarkers of O3I in two randomized placebo-controlled trials. An untargeted lipidomic data workflow using a subgroup analysis of serum extracts from sunflower oil versus high-dose fish oil (FO)-supplemented participants revealed that ingested n3-LCPUFAs were primarily distributed as their phosphatidylcholines (PCs) relative to other PL classes. In both high-dose FO (5.0 g/day) and EPA-only trials (3.0 g/day), PC (16:0_20:5) was the most responsive PL, whereas PC (16:0_22:6) was selective to DHA-only supplementation. We also demonstrated that the sum concentration of both these PCs in fasting serum or plasma samples was positively correlated to the O3I following FO (r = 0.708, P = 1.02 × 10−11, n = 69) and EPA- or DHA-only supplementation (r = 0.768, P = 1.01 × 10−33, n = 167). Overall, DHA was more effective in improving the O3I (ΔO3I = 4.90 ± 1.33%) compared to EPA (ΔO3I = 2.99 ± 1.19%) in young Canadian adults who had a poor nutritional status with an O3I (3.50 ± 0.68%) at baseline. Our method enables the rapid assessment of the O3I by directly measuring two circulating PC species in small volumes of blood, which may facilitate screening applications for population and precision health.

Long-chain omega-3 polyunsaturated fatty acids are reduced in neonates with substantial brain injury undergoing therapeutic hypothermia after hypoxic-ischemic encephalopathy.

Hypoxic-ischemic encephalopathy (HIE) is a major cause of neonatal morbidity and mortality. Although therapeutic hypothermia is an effective treatment, substantial chronic neurological impairment often persists. The long-chain omega-3 polyunsaturated fatty acids (PUFAs), docosahexaenoic (DHA) and eicosapentaenoic (EPA) acids, offer therapeutic potential in the post-acute phase. To understand how PUFAs are affected by HIE and therapeutic hypothermia we quantified for the first time the effects of HIE and therapeutic hypothermia on blood PUFA levels and lipid peroxidation. In a cross-sectional approach, blood samples from newborns with moderate to severe HIE, who underwent therapeutic hypothermia (sHIE group) were compared to samples from newborns with mild HIE, who did not receive therapeutic hypothermia, and controls. The sHIE group was stratified into cerebral MRI predictive of good (n = 10), or poor outcomes (n = 10; nine developed cerebral palsy). Cell pellets were analyzed for fatty acid content, and plasma for lipid peroxidation products, thiobarbituric acid reactive substances and 4-hydroxy-2-nonenal. Omega-3 Index (% DHA + EPA) was similar between control and HIE groups; however, with therapeutic hypothermia there were significantly lower levels in poor vs. good prognosis sHIE groups. Estimated Δ-6 desaturase activity was significantly lower in sHIE compared to mild HIE and control groups, and linoleic acid significantly increased in the sHIE group with good prognosis. Reduced long-chain omega-3 PUFAs was associated with poor outcome after HIE and therapeutic hypothermia, potentially due to decreased biosynthesis and tissue incorporation. We speculate a potential role for long-chain omega-3 PUFA interventions in addition to existing treatments to improve neurologic outcomes in sHIE.

Higher docosahexaenoic acid levels lower the protective impact of eicosapentaenoic acid on long-term major cardiovascular events.

Long-chain omega-3 polyunsaturated fatty acids (OM3 PUFA) are commonly used for cardiovascular disease prevention. High-dose eicosapentaenoic acid (EPA) is reported to reduce major adverse cardiovascular events (MACE); however, a combined EPA and docosahexaenoic acid (DHA) supplementation has not been proven to do so. This study aimed to evaluate the potential interaction between EPA and DHA levels on long-term MACE. We studied a cohort of 987 randomly selected subjects enrolled in the INSPIRE biobank registry who underwent coronary angiography. We used rapid throughput liquid chromatography-mass spectrometry to quantify the EPA and DHA plasma levels and examined their impact unadjusted, adjusted for one another, and fully adjusted for comorbidities, EPA + DHA, and the EPA/DHA ratio on long-term (10-year) MACE (all-cause death, myocardial infarction, stroke, heart failure hospitalization). The average subject age was 61.5 ± 12.2 years, 57% were male, 41% were obese, 42% had severe coronary artery disease (CAD), and 311 (31.5%) had a MACE. The 10-year MACE unadjusted hazard ratio (HR) for the highest (fourth) vs. lowest (first) quartile (Q) of EPA was HR = 0.48 (95% CI: 0.35, 0.67). The adjustment for DHA changed the HR to 0.30 (CI: 0.19, 0.49), and an additional adjustment for baseline differences changed the HR to 0.36 (CI: 0.22, 0.58). Conversely, unadjusted DHA did not significantly predict MACE, but adjustment for EPA resulted in a 1.81-fold higher risk of MACE (CI: 1.14, 2.90) for Q4 vs. Q1. However, after the adjustment for baseline differences, the risk of MACE was not significant for DHA (HR = 1.37; CI: 0.85, 2.20). An EPA/DHA ratio ≥1 resulted in a lower rate of 10-year MACE outcomes (27% vs. 37%, adjusted p-value = 0.013). Higher levels of EPA, but not DHA, are associated with a lower risk of MACE. When combined with EPA, higher DHA blunts the benefit of EPA and is associated with a higher risk of MACE in the presence of low EPA. These findings can help explain the discrepant results of EPA-only and EPA/DHA mixed clinical supplementation trials.

Evaluating Alternative and Sustainable Food Resources: A Review of the Nutritional Composition of Myctophid Fishes

Additional and alternative sustainable food resources are needed as the global human population increases. Marine fishes have long provided essential nutrients, such as omega-3 long-chain (≥C20) polyunsaturated fatty acids (n-3 LC-PUFA), protein, and vitamins to meet human dietary requirements and feed for agricultural production. Many current commercial fish stocks are depleted or fully exploited, but oceanic mesopelagic fishes, particularly the myctophids (lanternfishes), represent a potentially very large and unfished resource. This review analysed the literature on nutritional and biochemical compositions of myctophids as a first step towards understanding the health benefits and risks of consuming them. We found that myctophids have high levels of protein (11–23% wet weight, WW) and variable lipid content (0.5–26% WW). In most species, desirable triacylglycerols or phospholipids dominated over less-desirable wax esters, and most have abundant amounts of health-promoting n-3 LC-PUFA, such as DHA and EPA. Myctophids have low levels of heavy metals and persistent organic pollutants. Most nutritional information is available for species from the Pacific and Southern Oceans and for the genera Benthosema, Electrona, and Diaphus. Myctophids generally possess favourable nutritional profiles, but major gaps in knowledge regarding their stock assessment, ecology and the economic viability for their harvest are barriers to developing sustainable fisheries.
